Useful Tools / Techniques

  • ntwo-dimensional electrophoresis
  • ncomparison of two gels made of two different proteomes
  • npurify a particular spot on gel --> digest with protease--> mass spectrometry --> acquire amino acid composition to support identification of genes

Hybridization analysis

  • ncan detect not only expression patterns but also changes in expression patterns under two different conditions

 

Microarray (cDNA) Analysis

  • n6000 genes of yeast can be spotted onto glass slides in arrays of 80 spots X 80 spots
  • nmRNA extraction ( cDNA synthesis and fluorescence labeling ( assess hybridization via microarrays and  confocal microscopy)

DNA Chips

  • noligonucleotides are synthesized directly on the surface of the "chip"
  • ndensity of 1 million per cm2 (much higher) - great throughput for screening numerous samples

  • Proteome the protein content of a cell (the entire collection of proteins in a cell)
